1. PRINCIPLE OF METHOD An anion exchange column is used for the separation of the carbohydrates. The separation
mechanism of these columns relies on the fact that carbohydrates are complex alcohols that can be ionised at high pH, The acidity of each hydroxyl group depends on neighbouring groups and different
degrees of dissociation at certain pH values. With an eluant at the appropriate pH, many sugars can be separated rapidly by high performance ion chromatography. As the ionisation of sugars is pH
sensitive, these columns are notably more selective than the gel-based cation exchange columns.
